Estimation of various parameters of blood chemistry of albino mice on exposure to oral anti Diabetic Drug, Pioglitazone

Abstract : Pioglitazone Hydrolchloride (Piozone) is an oral antidiabetic agent belonging to the class of Thiazolidinediones (Glitazone) Pioglitazone is used in the treatment of type II Diabetes Mellitus and acts primarily by decreasing insulin resistance. Drugs of this newer class of antihyper-glycemic agents, Thiazolidinediones sensitize peripheral tissues to insulin. The present study attempts to estimate the biochemical parameters like Blood Glucose, Liver Function Test Profile, Kidney Function Test Profile and Lipid Test Profile of the albino mice after administration of different doses of Pioglitazone through drinking water. Estimation has been done after four and eight weeks of treatment. Doses of pioglitazone used in the present investigation (50, 100, and 200 ppm) are well tolerable by the mice upto 10 weeks duration. With the decreasing dose of piolitazone glucose level decrease significantly in both 4 weeks and 8 weeks treatment in comparison to normal control. SGOT level increase with increasing dose but does not exceed normal control level in 4 weeks treatment but at 8 weeks SCOT level increases with increasing dose and has exceeded the normal control level. No remarkable changes were observed for Alkaline Phosphatase and Albumin and Globulin. Kidney Function Test Profile has shown a slight decrease in the Blood urea on the increasing the dose whereas Total protein has decreased. Lipid test Profile also showed some remarkable changes on different doses of Pioglitazone.
